Abstract

Background. Prevention of dental caries is expensive and only to prevent S. mutans attachment to the teeth, but it can’t increase of the immune response. Caries prevention, S. mutans must be attached to salivary leukocytes and then destroyed it. U. moluccensis scales that contain various amino acids, Calcium, Phosphorus, flavonoids are thought to increase S. mutans adhesion in salivary leukocytes. This study aimed to analyze the effect of fish scales of U. moluccensis on S. mutans adhesion activity on salivary leukocytes. Methods. About 5ml of saliva was collected in 50ml sterile Falcon® tubes placed on ice. Furthermore saliva was added with dPBS and Ficoll hipaque, centrifugation and suspended in RPMI 1640 medium. The cells were placed in 24-well microtiter plate, incubation and washed, treated according to groups. Controle Group: Untreated Leukocytes, S. mutans Group: leukocytes + S. mutans. Group fish scales: fish scales + leukocytes + S. mutans. Adhesion was stained with Giemza. Cells were counted under a light microscope with magnification 400 times per 100 cells. The data obtained were analyzed using Anova followed by LSD test. Results. The S. mutans adhesion activity in salivary leukocytes were treated with more fish scales than the group that was not given fish scales. This may be due to the content which function as immunomodulators, which can increase recognition leukocytes to S. mutans. Conclusions. U. moluccensis scales can increase adhesion to S. mutans in salivary leukocytes, so that it can be used as dental material for prevent dental caries.
