Triplex-based Transcriptional Regulation in Chinese Hamster Ovarian Cells

ssDNA oligos have been shown to form triplex structures with dsDNA in vitro. we attempt to demonstrate the formation of these structures in vivo, in CHO cells. to do so, we designed ssDNA sequences (TFOs - triplex-forming oligos) that target a specific dsDNA motif (TTS - triplex target site). we present results showing the regulatory effect of these oligos on a synthetic construct.
