The talk will focus on the emerging application of mRNA-based technologies in T-cell immunotherapy. I will present data on developing a T-cell-optimized mRNA backbone that enables precise and differential gene expression. In addition, I will highlight strategies to improve T-cell function by delivering mRNAs that promote a stem-like, memory-associated phenotype to counteract exhaustion. Lastly, I will discuss the design of novel nanoparticle systems for targeted in vivo delivery of therapeutic mRNAs to human T-cells. Together, these approaches provide new avenues for T-cell reprogramming and mRNA-based therapies in cancer treatment.
