T-cells are important component of the anti-tumor response. We will present recent advances in enhancing engineered T-cell efficacy within the hostile tumor microenvironment. Strategies include metabolic reprogramming and signaling redirection to improve T-cell persistence, activation, and cytotoxicity. We will also focus on overcoming immunosuppression by enabling T cells to resist or exploit suppressive cytokine signaling. These integrated approaches point to novel design principles for next-generation adoptive cell therapies in solid tumors.
