GENERATION AND CHARACTERIZATION OF ANTIBODY TO GLOBO H AND SSEA-3

Monoclonal antibodies (Abs) therapies against tumor-associate glycans (TAG) have the potentiality in immunotherapy. Globo H (GH) is a hexasaccharide overexpressed on a variety of cancer cells. GH and stage-specific embryonic antigen 3 (SSEA-3) also reported specifically overexpressed on breast cancer cells and breast cancer stem cells. In this study, we used GH-DT/C34 as vaccine and screen antiserum by glycan array. Glycan microarray showed that GH-DT could elicit high titers of IgG antibodies to globo H and SSEA-3, the information was helping us for hybridoma production. From mice immunized with a GH-DT/C34 synthetic conjugate, IgG1 monoclonal antibody (mAb MC-27) was derived. Not only did mAb MC-27 recognized GH and SSEA-3 but also in human tumor cells MCF-7 which expressed GH.
