Next-Generation CAR-T Cell Therapies for Multiple Myeloma: Advancing Innovation at Hadassah

Hadassah Medical Center, in collaboration with Bar Ilan University, has developed and implemented a first-in-Israel CAR-T cell therapy targeting BCMA, known as HBI0101, which has been administered to over 150 patients with relapsed/refractory multiple myeloma and AL amyloidosis. The treatment has shown impressive responses, including deep and rapid clinical remissions. However, relapse remains common, particularly in high-risk groups, such as patients with extramedullary disease or those previously exposed to anti-BCMA therapies. Comprehensive correlative analyses have revealed strong associations between the differentiation profile of infused CART cells and treatment outcomes, motivating the development of a new manufacturing platform aimed at enriching for early memory T cells prior to genetic engineering. This presentation will explore novel strategies to improve CAR-T efficacy and durability, including a newly developed manufacturing platform, resulting in a more potent and long-lasting anti-tumor response. These advancements represent a translational approach bridging laboratory insights and clinical application, aiming to define the next generation of CAR-T therapies in myeloma.
